Total plasma homocysteine level is an important predictor of cardiovascular risk and correlates with severity of coronary artery disease. Very recent prospective, double-blind, randomized trial showed decreased rate of coronary restenosis after lowering of plasma homocysteine levels. This new study indicates that elevated homocysteine is causal for cardivascular disease. Therefore it is essential that will be mass screening for homocysteine levels. A/C DIAGNOSTICS enzymatic homocysteine test is simple, rapid, automated, economic and therefore highly suited for mass screening. The assay is based on a highly specific recombinant enzyme homocysteine a,g-lyase (rHCYase) using a modified methylene blue determination of H2S as analyte for homocysteine.

Introduction Pyridoxal 5-phosphate (PLP), is the biologically active form of the vitamin B6. Clinical studies suggest that low levels of PLP are an independent risk factor of cardiovascular disease. However, PLP levels are not routinely diagnosed due to lack of homogeneous, non-radioactive assays. We have now developed a homogeneous, non-radiactive enzymatic PLP assay using the PLP-dependent recombinant enzyme homocysteine a,g-lyase (rHCYase). The new PLP assay is the first homogeneous, non-radiactive B6 diagnostic applicable on chemistry autoanalyzers and should be of wide-based clinical use.
